Enclomiphene citrate is a non-steroidal selective estrogen receptor modulator (SERM) that acts as an antagonist at the estrogen receptors in the hypothalamus and pituitary gland. It is one of the two isomers of clomiphene citrate, better known under the brand name Clomid.
Compared to the racemic mixture of clomiphene, which is made up from both the enclomiphene and zuclomiphene isomers, enclomiphene has a shorter half-life and doesn’t have the unwanted side-effects associated with Clomid.
- Enclomiphene is the isomer that blocks estrogen receptors and stimulates testosterone production.
- Zuclomiphene is longer-acting, displays mild estrogenic activity, and may contribute to side effects.
Bodybuilders and athletes have widely adopted this compound for post-cycle therapy after a cycle of selective androgen receptor modulators (SARMs) and anabolic androgenic steroids (AAS). Blocking estrogen receptors in these tissues prevents negative feedback inhibition of gonadotropin release, leading to increased production of luteinizing hormone (LH) and follicle-stimulating hormone (FSH).
Because its action focuses more on enhancing FSH activity than LH activity, there is less feedback suppression than traditional SERMs or aromatase inhibitors (AIs).

How does enclomiphene work?
Enclomiphene Citrate has been scientifically proven to improve hormone levels while preserving sperm production. Because of this, it’s much safer, easier, and more ‘user-friendly’ than testosterone therapy, which can lead to serious health issues, amongst many other complications.
While most SERMs (selective estrogen receptor modulators) block some estrogens in the body, Enclo blocks all estrogen receptors throughout the body. It then stimulates the release of follicle-stimulating hormones by antagonising the negative feedback on the gonadotropin-releasing hormone (GnRH).
Mechanism of action
- Blocks estrogen in hypothalamic and pituitary tissues
- Elevates LH and FSH production from the pituitary gland
- Increased GnRH secretion from the hypothalamus
- Stimulates endogenous testosterone production
- Preserves fertility by maintaining FSH levels
Enclomiphene demonstrates the ability to restore testosterone production without the introduction of exogenous androgens, making it an effective compound for the purpose of post-cycle therapy.
Enclomiphene benefits
Raises Endogenous Testosterone
Enclomiphene stimulates LH and FSH, which signal the testes to make more testosterone. This mechanism helps restore hormonal balance without suppressing the body’s natural production. It does not harm sperm count, unlike TRT, resulting in a positive effect on fertility.
Preserves Fertility
Because LH and FSH remain active, sperm production is maintained. A 2023 head-to-head study of enclomiphene vs. clomiphene showed a significant rise in total motile sperm count with enclomiphene.
Potential side effects
Reported side effects are generally mild and include headache, nausea, or digestive discomfort. Some men experience transient increases in estradiol.
Enclomiphene dosage
Dosages used in research
Information about the most optimal enclomiphene dose comes from both human trials and preclinical research. What follows is an overview of the dosages used in research studies.
- 12.5 mg to 25 mg administered in a single daily dose in clinical studies
- 25 mg daily used in male hypogonadism research
- 6.25 mg to 12.5 mg administered during a cycle of SARMs to prevent suppression of natural testosterone production
Dosages used in post-cycle therapy
The enclomiphene protocol for recovery after a cycle of SARMs or anabolic steroids is based on both clinical data and a vast amount of anecdotal evidence from user reports.
- 12.5 mg to 25 mg per day for a period of 4 weeks, immediately taken after finishing a cycle of suppressive agents
Legal status
Enclomiphene has not been approved by the FDA or EMA for human consumption or therapeutic use. While clomifene, better known as Clomid, is a prescription drug, the eclomiphene isomer is not approved as a licensed medicine in the EU. It can therefore not be legally prescribed or sold as a supplement.
Enclomiphene is banned by the World Anti-Doping Agency (WADA) and remains prohibited in all levels of competitive athletics.
